One was the nickname of the leader of the USS mission to secure The Hive and shut down the Red Queen. His real name was James Shade. He was highly respected by Major Timothy Cain for his silent, businesslike, demeanour, keeping his cool under fire, and his unquestioning understanding of the chain of command his as well orders. He was a long-time friend of Cain and may have served with him during Desert Storm.

The evident incompetence of the Raccoon City Police Department by not having Matt Addison on their file since he was a new entry greatly angered him, since he enjoyed order despite being a soldier. He filled in Alice and Spence Parks on the situation and what they were doing in the mansion after the Red Queen gave them amnesia. He greatly impressed them with his leadership skills.

Later, he went with Olga Danilova, Alfonso Warner, and Vance Drew into the Red Queen's chamber but was cut off. When the Red Queen activated its laser grid, he brought Drew to the ground in an attempt to save him but Drew rose his arm instinctively and his hand got chopped off. He lost his cool briefly (a very rare event) when Drew started to lose his consciousness from shock and Olga was killed, keeping Drew from medical attention. Despite his efforts, he could not save Drew. Soon, Warner was dead and One was the only survivor left. A fairly high laser flew towards him, and he jumped up, grabbed hold of the piping and lifted himself up, the laser barely missing (managing to chop his knife in two). The Red Queen made the laser grid into an unstoppable diagonal chain-link grid which One could not dodge. It cut him apart into tiny pieces and killed him. Moments after his death, Chad Kaplan disabled the grid, with another metre before reaching the end, which could have saved him had he gone there.

He was played by Colin Salmon.
